CS/DS sulfatases are a type of enzymes that can specifically hydrolyze and remove the sulfate groups at a specific positions of CS/DS chain. They are important tools for studying the structure-activity relationship of CS/DS, including GalNAc-4-O-sulfatases, GalNAc-6-O-sulfatases and GlcUA/IdoUA-2-O-sulfatases, etc. Similar to other GAG-degrading enzymes, GAG sulfatases can also be divided into two categories: endosulfatase, which only removes the sulfate group at the end of the CS/DS chain, and exosulfatase, which can act on not only the sulfate groups at the end of CS/DS chain but also the internal sulfate groups of CS/DS chain. Therefore, CS/DS endosulfatases can be used as powerful tools to more effectively tailor and edit the sulfation patterns of CS/DS, thereby to regulate their biological functions.
